The verb "to die" can be interpreted in different ways.

It can mean "stop living", "become extinct" but also "be forgotten".

A human being does not die only when his/her hearth stops, when he/she can no longer breath.

What Shakespeare is trying to communicate in the sentence is that a person who is cowardly dies many times before really dying. But what does acting cowardly mean?

In my opinion, a person acts cowardly when he/she does not face a situation, when he/she runs away from a difficult situation, when he/she does not  choose a way of acting for their ideas or values but just because it is the easiest way to take.

Every situation you run away from, every escape you make, every decision you didn't take is like dying a little inside.

On the contrary, fearless people, who have the bravery of taking their decision, who knows the consequences their decision may have, but that however follow their ideas will die only once. They will die just when his/her heart will stop, fear will not stop them.
